INTENDED USE
Monoclonal Anti-D blend - testserum is produced from cell culture supernatants of heterohybridoma
cell lines. One cell line is secreting an antibody of IgG-type and the other an antibody of
IgM-type, that both react specific with the corresponding antigen. The testserum is used to determine
whether human red blood cells possess or lack the corresponding blood group antigen D. This
testserum reacts negatively with erythrocytes of category DVI with the IgM-antibody and positive
with the IgG-antibody in indirect Coombs-test. The testserum is intended be used by qualified and
technical personnel only.
PRINCIPLE OF PROCEDURE
The procedures used with this reagent are based on the principle of agglutination. Normal human
erythrocytes, possessing the corresponding antigen, will agglutinate in the presence of the specific
antibody directed towards the antigen.
REAGENTS
The listed reagent contains antibodies of the following cell clones:
Anti-D blend (monoclonal, human IgG + IgM, clones: MS-26; TH-28)
The reagent contains <0.1% (w/v) sodium azide as preservative. Additionally the reagent is
prepared of active antibody, sodium chloride, macromolecules and bovine albumin.
WARNING
This reagent was prepared from supernatants of cell cultures. As a biological product it should be
looked upon as potentially infectious because of never complete exclusion of danger through
exitants of disease. The reagent contains sodium azide, that may be toxic and may react with lead
or copper to form highly explosive salts. Because of these reasons this reagent should be handled
with proper care.
STORAGE REQUIREMENT
Store at 2 to 8°C. May be at room temperature (15 to 30°C) while in use. In principle, store and use
the reagents to indicated expiry date only.At keeping of storage conditions after bottle opening keep performance data to a validity expiration date.
REMARKS
1. Strength of positive reactions also depends on age of used blood
2. With each testing positive and negative controls should be performed.
3. Inappropriate storage impairs efficacy of the reagent.
4. Centrifugation highly different from appointed relative centrifugal force may lead to false results.
5. Blood samples to be tested should be used as soon as possible. If a delay in testing occurs,
samples should be stored at 2 to 8°C. Blood drawn into sodium citrate or EDTA should be
tested within 14 days. Blood obtained by finger puncture may be tested directly by the slide
method but, to avoid clotting, blood collected in this manner should be mixed quickly with the
reagent.
6. The procedures identified below are for manual testing only. When using automated or semiautomated
instruments, follow the procedures that are contained in the operator’s manual
provided by the device manufacturer. Laboratories must follow approved validation procedures
to demonstrate compatibility of this product on automated systems.
7. For usage of this testsera all effective national laws, directives and guidelines have to be
observed, in Germany especially the „RichtlinienzurGewinnung von Blut und Blutbestandteilen
undzurAnwendung von Blutprodukten (Hämotherapie)“.

There is no preparation of the reagents required. Use reagents directly from the vials.
PROCEDURE

at Tube Centrifugation Method: Test tubes, 10 x 75 mm or 12 x 75 mm; Pipettes designed to
deliver approximately 100 μL; Centrifuge; Isotonic saline (with 0,85 - 0,9% sodium chloride)
at Card Method: Cards (DiaMed ID-Card “NaCl, enzyme test and cold agglutinins”,
BIO-RAD „Scangel neutral“, Ortho Clinical Diagnostics „Biovue System Reverse Diluent” or
Grifols “DG Gel Neutral); microliter pipette; Card Centrifuge; card specific diluent.
atMicroplate Method: microplate with 96 U-wells; microliter pipette; centrifuge; shaker;
Isotonic saline (with 0,85 - 0,9% sodium chloride)
Test procedure
Slide Method
1. Use erythrocyte sediment or whole blood only.
2. Place one drop (approximately 50 μL) of appropriate reagent on a glass slide.
3. Using a Pasteur pipette add one drop erythrocyte sediment or whole blood (approximately
50 μL) to the glass slide.
4. Mix the erythrocytes with reagent well with a stick and spread to a circle (diameter 2 cm)
5. By slightly rotating the slide, check for agglutination within 1 minute (reaction starts within
seconds). Unspecific reactions might appear due to drying of the reaction-formation or if the
slide is heated.
Tube Centrifugation Method
1. Use a 2% to 5% suspension of red blood cells in isotonic saline (cells washed one time or up to
three times with isotonic saline) only.
2. Add 100 μL (alternative: one drop = approximately 50 μL) of appropriate reagent to each tube.
3. Add 100 μL (alternative: one drop = approximately 50 μL) of appropriate cell suspension to
each tube.
4. Mix well by slightly shaking.
5. Incubate tube at room temperature (15 to 30 °C) for 1 - 15 min.
6. Centrifugation of tube for 1 minute at 2.000 rpm (approximately 800-1.000 x g).
7. Gently resuspend the red cells and check macroscopically for agglutination within 3 minutes.
Document the result.
Indirect Coombs-test
8. Repeat step 1.-4. with fresh material or continue after reading of result directly.
9. Incubate tube at 37 °C for 30 min.
10. Wash red cells 3 times with (cold) isotonic saline.
11. Add 100 μL Anti-Human-Globulin-serum (Coombs-serum) to each tube
12. Centrifugation of tube for 1 min at 1.000 rpm (approximately 180-270 x g).
13. Gently resuspend the red cells and check macroscopically for agglutination within 3 minutes.
Document the result.
Card Method
1. Use 0,8 % suspension of red blood cells in card specific diluent.
2. Add 50 μL of appropriate cell suspension to each micro tube.
3. Add 25 μL of appropriate testserum to each micro tube (alternative one drop). If automated
tech-niques are used add 10 μL testserum. The use of 10 μL testserum is possible in hand
method too.
4. Centrifugation of card in appropriate card centrifuge after latest 30 minutes.
5. Check macroscopically for agglutination within 30 minutes. Document the result.
Microplate Method
1. Use 2% suspension of red blood cells in isotonic saline (cells may be washed one time or up to
three times with isotonic saline).
2. Add 50 μL of appropriate cell suspension to each well.
3. Add 50 μL of appropriate testserum to each well.
4. To mix both 30 seconds on shaker with maximum speed.
5. Centrifugation of plate in appropriate centrifuge for 30 seconds at 400 x g.
6. For 30 seconds on shaker with medium speed, check macroscopically for agglutination.
Document the result.

НАЗНАЧЕНИЕ
Моноклональная агглютинирующая тест-сыворотка Анти-Dblend изготовлена из надосадочных жидкостей клеточных линий гетерогибридомы. Одна клеточная линия продуцирует антитела класса IgG, другая антитела класса IgM, оба класса специфично взаимодействуют с соответствующими антигенами. Тест-сыворотка определяет наличие или отсутствие антигена D. Даннаятест-сыворотка дает отрицательную реакцию с антигеном DVIc антителами IgM и положительную с антителами IgG в непрямом антиглобулиновом тесте. Тест-сыворотка может использоваться только квалифицированным персоналом.
ПРИНЦИП ИССЛЕДОВАНИЯ
Исследование с использованием данных реагентов основано на принципе агглютинации. Нормальные человеческие эритроциты, покрытые соответствующими антигенами, реагируют с присутствующим специфическим антителом, направленным против антигена.
РЕАГЕНТЫ
Данный реагент содержат антитела следующих клеточных клонов:

Все реагенты содержат <0,1 % азида натрия в качестве консерванта. Также реагенты содержат активные антитела, хлорид натрия, макромолекулы и альбумин бычьей крови.
ПРЕДУПРЕЖДЕНИЕ
Данные реагенты изготовлены из надосадочных жидкостей клеточных культур.Данный биологический продукт следует рассматривать в качестве потенциально инфицированного, не исключайте опасности заражения вследствие наличия возбудителей заболевания. Реагент содержит азид натрия, который может быть токсичным и реагировать со свинцом или медью, образуя взрывоопасные соединения.Вследствие этого эти реагенты должны использоваться с большой осторожностью.
УСЛОВИЯ ХРАНЕНИЯ
Хранить при температуре 2 – 8°С. В процессе использования можно хранить при комнатной температуре (15 – 30°С). Вообще, хранение и использование реагентов зависит от даты истечения срока годности.При соблюдении условий хранения после вскрытия флакона сохраняют рабочие характеристики до даты истечения срока годности.
ПРИМЕЧАНИЯ
1. Сила положительной реакции также зависит от возраста используемой крови
2. В каждом исследовании должен выполняться положительный и отрицательный контроль.
3. Ненадлежащее хранение способствует снижению активности реагента.
4. Сила центрифугирования, отличная от указанной, может привести к неправильным результатам.
5. Образцы крови по возможности должны быть исследованы как можно быстрее. Если исследования задерживаются, образцы должны храниться при температуре 2 – 8°С. Кровь с цитратом натрия или ЭДТА должна исследоваться в течение 14 дней. Взятая из пальца кровь может быть исследована на плоскости, но, во избежание образования сгустков, такая кровь должна быть как можно быстрее смешана с реагентом.
6. Исследования, описанные ниже, подходят только для ручного метода. При использовании автоматического и полуавтоматического оборудования следуйте предоставленному производителем Руководству по использованию инструмента. Сотрудники лабораторий должны следовать установленным системам оценки исследований, чтобы удостовериться в идентичности результатов, полученных ручным и автоматическим методом.
7. Перед использованием данной тест-сыворотки нужно ознакомиться с соответствующими национальными законами (приказами, директивами, инструкциями).

ИССЛЕДОВАНИЕ
Реагент не обеспечен материалом, дополнительно требуются:
Для метода на плоскости: Планшет, пипетка Пастера, Палочки для смешивания.
Для метода центрифугирования в пробирке: Пробирки 10 х 75 или 12 х 75 мм, пипетки с возможностью дозирования 100 мкл, центрифуга, изотонический раствор (0,85 – 0,9 % натрия хлорида).
Для гелевого метода: Гелевые карты (DiaMedID-Card «NaCl, ферментный тест и холодовая агглютинация», BIO-RAD «Scangelneutral», Grifols «DGGelNeutral»или OrthoClinicalDiagnostics «BiovueSystemReverseDiluent», Пипетка, позволяющая дозировать микролитры, Центрифуга для гелевых карт, раствор для гелевых карт).
Для работы в микроплатах: микроплата с 96 U-образными лунками, Пипетка, позволяющая дозировать микролитры, Центрифуга, Шейкер, изотонический раствор (0,85 – 0,9 % натрия хлорида).
Алгоритм исследования
Метод на плоскости
1. Используйте осадок эритроцитов или цельную кровь.
2. Поместите одну каплю (приблизительно 50 мкл) соответствующего реагента на планшет.
3. Используя пипетку Пастера, добавьте одну каплю осадка эритроцитов или цельной крови (приблизительно 50 мкл) на планшет.
4. Тщательно смешайте эритроциты с реагентом и сформируйте круг (диаметром 2 см).
5. Осторожно вращая планшет, проверьте агглютинацию в течение 1 минуты (реакция начнется в течение нескольких секунд). Неспецифические реакции могут возникнуть при высыхании реактивной формации или нагревания.
